it varies. My current lot start showing a spot about 10-20 days from birth, where my old lot seemed to never loose theirs. It starts when she becomes pregnant and gets darker closer to time, so when its nearly black, she's ready to go at any day and you can expect some little guppies :)
If there are male gupppies around, then u should see gravid spot most of the time as female guppies are pregnant again as soon as they pop! btw, females store sperm within them, which is good for several rounds of pregnancies.
when they are with males they will almost constantly have one.. but it depends on how stressed they are. females can reject a pregnancy if the conditions are not right - kind of like a DIY abortion. so if your water quality is not good they may never become pregnant
